Foi um dos portugueses que, de Malaca, se dirigiram à China, sendo o primeiro a chegar ao Sul da China, em 1513. A esta visita seguiu-se o estabelecimento de algumas feitorias portuguesas na província de Cantão, onde mais tarde se viria a estabelecer o entreposto de Macau. De acordo com os registos disponíveis, foi o primeiro europeu a alcançar e visitar o território que actualmente é Hong Kong.

Possuía um Junco com o qual se dedicava ao comércio entre Malaca e Cantão, juntamente com Simão de Andrade e Rafael Perestrelo, pioneiros desse comércio, considerado ilegal pelos Chineses.

Participou de uma guerra contra o sultão de Bintão, capitaneando uma galé na Armada Portuguesa. Com a abordagem de Tamang (Cantão), apesar da oposição do "Itau" (mandarim local), conseguiu estabelecer-se em uma praia na ilha de Sanchoão, onde ergueu uma cabana que servia de refúgio aos comerciantes clandestinos e onde, para se achar como em terra portuguesa, fizera assentar um padrão.

Passou assim a ser considerado como feitor português de Tamang, continuando, no seu Junco, a navegar pelas Molucas. Nestas águas veio a ser atacado pelos indígenas de Ternate, vindo a ser gravemente ferido. Veio falecer na sua cabana, pedindo que fosse enterrado junto ao padrão que fizera erigir.

Há informações acerca de outro Jorge Álvares (um filho, um homónimo?), um rico mercador português que, em 1544, foi ao Japão com Fernão Mendes Pinto e que escreveu "Informação do Japão", a pedido de São Francisco Xavier. Auxiliou ainda este religioso tendo conduzido, da China, o seu converso japonês de nome Anger. Em 1552 o religioso aportou à ilha de Sanchoão gravemente enfermo, tendo sido acolhido por Jorge Álvares na sua cabana. Jorge Álvares (died July 8, 1521) is credited as the first Portuguese explorer to have reached China and Hong Kong. The Fundação Jorge Álvares (Jorge Álvares Foundation), founded by Vasco Joaquim Rocha Vieira prior to the handover of Macau, got its name for also having reached there.

In May 1513 Álvares sailed under the Portuguese Malacca captain, Rui de Brito Patalim in a junk from Pegu. The expedition was accompanied by 5 other junks. Álvares himself was accompanied by 2 other Portuguese mariners.

Álvares made first contact on Asian soil in Guangdong, Southern China in May 1513. Upon landing, he raised a Padrão from the king of Portugal, where they landed on Lintin Island in the Pearl River estuary. Based on information from their captain, they were to hope to find trade. Soon after this, the Viceroy of the Estado da India dispatched Rafael Perestrello—a cousin of Christopher Columbus— to seek trade relations with the Chinese. In a ship from Malacca, Rafael landed on the southern shores of Guangdong later that year in 1513, being the first to actually land on the coast of mainland China.

Álvares later joined the venture of establishing the settlements in Tuen Mun, Hong Kong around 1513 to 1514. This visit was followed by the establishment of a number of Portuguese trading centres in the area, which were eventually consolidated in Macau. In 1517 the Portuguese settlers were in a battle with Imperial Chinese army troops in the region.
